Description

Description: This assay employs a two-site sandwich ELISA to quantitate INO80B in samples. An antibody specific for INO80B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any INO80B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for INO80B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of INO80B bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: In Saccharomyces cerevisiae, orthologs of Tip49a and Tip49b, called Rvb1 and Rvb2, respectively, are subunits of two distinct ATP-dependent chromatin remodeling complexes, SWR1 and INO80. The mammalian Tip49a and Tip49b proteins are integral subunits of a chromatin remodeling complex bearing striking similarities to the S. cerevisiae SWR1 complex.A new mammalian Tip49a- and Tip49b-containing ATP-dependent chromatin remodeling complex, which includes orthologs of 8 of the 15 subunits of the S. cerevisiae INO80 chromatin remodeling complex as well as at least five additional subunits unique to the human INO80 (hINO80) complex.
